摘 要:目的研究银杏叶提取物(EGB)对紫外线所致体外培养兔晶状体氧化损伤的保护作用。方法将体外培养兔晶状体分为正常培养组,紫外线照射对照组(波长260~365 nm,强度400 W/cm2,时间30 min),照射前EGB保护组,照射中EGB保护组,照射后EGB保护组。照射前后各培养24 h,观察各组晶状体混浊情况,并测定晶状体中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、活性氧物质(ROS)以及丙二醛(MDA)的含量。结果紫外线照射组晶状体全部混浊,SOD、GSH-Px、CAT酶活性降低,ROS及MDA含量升高。照射前及照射后EGB保护组晶体混浊率较对照组明显降低(P<0.05)。各EGB保护组抗氧化酶活性均较对照组升高,ROS及MDA含量降低,其中照射前及照射后EGB保护组与对照组相比,差异有统计学意义(P<0.05)。结论紫外线照射引起兔晶状体氧化损伤,EGB可作为一种抗氧化剂减轻这种氧化损伤。Objective To evaluate the effect of extract of Ginkgo biloba(EGB) on protecting lens against UV radiation-induced oxidative stress.Methods rabbit's lens cultured in vitro were divided into 5 groups:normal group,UV irradiated group(260~365 nm,400 W/cm2,30 min),EGB treatment before irradiation group,EGB tretment during irradiation group,EGB tretment after irradiation group.The transparency of all lens was examined.At the same time,the levels of SOD,GSH-Px,CAT and ROS,MDA were measured respectively.Results All lens of UV group developed into cataract,SOD,GSH-Px,CAT activities significantly decreased,ROS and MDA contents significantly elevated.Lower lens of EGB tretment before and after irradiation group developed into cataract(P〈0.05).The activities of antioxidant enzymes elevated in ECB groups,ROS and MDA contents decreased.Differencese are markedly in EGB tretment before and after irradiation group,compared with UV group(P〈0.05).Conclusion As a preventive antioxidant,EGB can protect rabbit's lens against UV irradiation-induced oxidative stress.
