摘  要:[目的]优化和筛选小麦洛夫林10悬浮细胞系培养的条件。[方法]以成熟胚为外植体,探讨了附加不同浓度KT对愈伤组织诱导和继代培养的影响,并通过调整2,4-D、KT和蔗糖的浓度及初始接种量对悬浮细胞系的培养条件进行了优化筛选。[结果]小麦洛夫林10愈伤组织的诱导培养基中不宜加入KT;初始接种量为1.5 g,液体培养基中添加1.00~2.00 mg/L 2,4-D、0.05~0.10 mg/L KT和30.00g/L蔗糖,最有利于小麦洛夫林10悬浮细胞系的稳定。[结论]为以悬浮细胞系为材料深入开展小麦抗病机制的研究奠定了基础。[Objective] The aim was to optimize and screen culture conditions of cell suspension of wheat lovrin 10.[Method] Taking mature embryos as explants,effects of different mediums with 2,4-D and KT on callus induction and subculture were studied,furthermore the culture conditions of cell suspension were optimized and screened by adjuesting the concentrations of 2,4-D,KT and sucrose and the intial inoculum.[Result] KT was not necessary in the culture medium of callus induced.The intial inoculum of the cell suspension was 1.5 g.1.00-2.00 mg/L 2,4-D,0.05-0.10 mg/L KT and 30.00 g/L sucrose were the effective concentration for stabilization of the cell suspension.[Conclusion] The research lays the foundation for further studying wheat resistant mechanism by taking cell suspension as material.
