摘  要:采用蛋白电泳和线粒体 DNA RFLP分析方法,研究了浙江七个地方品种猪及浙江野猪的遗传多样性状况⒚从分析的30 个遗传座位、99 个地方品种个体中发现 MDH,PEPB,6PGD,AK,ADA,PHI六个座位具有多态性,其中AK,MDH 多态座位在家猪中为首次发现⒚多态座位百分比和平均杂合度分别为P= 0200, H= 0043⒚用UPGMA 法对由基因频率所计算的标准遗传距离进行聚类分析,结果显示:浙江地方猪种的亲缘关系很近⒚在m tDNA RFLP水平,25 种限制性内切酶在27 个地方品种个体及3 个野猪个体中共检出30 种限制性态型,可归结成三种单倍型⒚各单倍型间平均遗传距离(P)为0005,群体平均多态度(π)为0000 8,处于遗传多样性贫乏范围⒚另外对金华Ⅲ系猪的研究表明金华Ⅲ系猪可能有两个母系起源⒚浙江省地方猪种遗传多样性贫乏。Protein electrophoresis and mtDNA RFLP were adopted to discover the genetic diversity of 7 native pig breeds and wild pig in Zhejiang Our preliminary results indicate that out of the 30 protein loci examined in the present experiment , their genotypes and gene frequencies were almost similar among 99 native pigs, only the ADA, AK, MDH, PEPB, 6PGD, PHI showed polymorphism The discovered polymorphism of AK and MDH had never been found in domestic pigs and wild pigs The percentage of polymorphical loci and mean herterozygosity are P =0 200 and H =0 043 respectively UPGMA dendrogram of native breeds(strains) was constructed by genetic distance From the phenogram, it is clear that the relationships among the native breeds are close On mtDNA RFLP, among the 27 native pigs and 3 wild pigs, 30 restriction morphs were discovered using 25 restriction endonucleases, which could be grouped into 3 haplotypes The average genetic distance ( ) was 0 005, and the value of average nucleotide diversity ( ) in pig population (including native pigs and wild pigs) was 0 000 8 The results suggest that the genetic diversity of pigs is remarkably scarce and the Jinhua strain Ⅲ pigs may have two female origins, and the Zhejiang native breeds may originate from a common ancester of wild pigs
