摘 要:目的探讨妇女宫颈上皮脱落细胞hTERC基因扩增情况和临床意义。方法采用荧光原位杂交技术(FISH)检测20例健康人及100例妇科疾病患者宫颈上皮脱落细胞hTERC基因扩增情况,确定hTERC基因扩增阳性阈值,研究临床病理诊断与hTERC基因扩增的关系。结果 FISH检测hTERC基因扩增阳性阈值为3.4%,CIN1患者FISH检测阳性率为8.8%(3/34例),CIN2患者为35.7%(5/14例),CIN3患者为77.8%(7/9例)以及宫颈癌患者为83.7%(36/43例)。健康对照组及CIN1者hTERC扩增发生机率显著低于CIN2及以上者(P<0.01)。结论 FISH可以成功地应用于妇女宫颈上皮脱落细胞hTERC基因扩增情况的研究;hTERC基因扩增与临床病理诊断密切相关。Objective To study the clinical value of hTERC status of cervical epithelial cells.Methods Cervical secretion samples from patients with cervical lesions and healthy women were detected by fluorescence in situ hybridization(FISH).hTERC amplification threshold and the relationship between hTERC and pathological findings were analyzed.Results The positive threshold of hTERC gene was 3.4%,and with 8.8%(3/34) of CIN1,35.7%(5/14cases) of CIN2,77.8%(7/9cases) of CIN3 and 83.7%(36/43 cases) of cervical carcinoma,respectively.hTERC amplified incidence of healthy and CIN1 groups were significantly lower than those of CIN2 and higher grades of cervical lesions(P0.01).Conclusions FISH has been successfully applied to detect hTERC gene in exfoliative cervical cells,and hTERC status is associated with clinical pathology diagnosis of cervical lesions.
