摘  要:目的:筛选小鼠血清中紫杉醇浓度测定的方法。方法:以Agilent C18柱为固定相,检测波长为227 nm,流速为1 mL.min-1,25℃条件下对流动相、内标、提取方法进行筛选。结果:确定以固相萃取方法提取样品,内标为地西泮和多西紫杉醇,流动相为磷酸水溶液-乙腈(50:50 V/V)以达到最佳的分离效果,测定线性范围在0.39~100.00μg.mL-1,回归方程为Y=13.325X+0.851 9(R2=0.999 3)。结论:筛选出的方法血清干扰少,样品测定周期短、快速、准确,为血清中紫杉醇浓度的测定提供了优选的方法。Objective: To screen a method for determining the concentration of paclitaxel in serum.Methods: The C18 column was used and the detection was carried out at 227 nm with UV detector.The mobile phase,internal standard and extraction method were optimized at 25 ℃.Results: Diazepam and taxotere was used as the internal standard,the solid phase extraction(SPE)were used to extract the sample,the mobile phase consisted of phosphoric acid: acetonitrile(50:50 V/V) delivered at a flow rate of 1 mL·min-1.The calibration curves were linear in the rang of 0.39~100.00 μg·mL-1 for paclitaxel.Regression equation:Y=13.325X+0.851 9(R2=0.999 3).Conclusion: This optimized method has less interference with serum.It's a rapid and accurate way to measure the concentration of paclitaxel in serum.
