针刺对慢性脑缺血大鼠Aβ蛋白影响的研究

摘　　要：目的观察针刺对脑组织内Aβ蛋白表达的变化,探讨针刺治疗慢性脑缺血的作用机制。方法采用大鼠分次永久性双侧颈总动脉阻塞(2VO)法制备慢性脑缺血大鼠模型并观察不同组别大鼠脑组织病理形态学的变化及脑组织内Aβ蛋白水平的变化。结果模型组大鼠神经元变性严重,而立即针刺组及针刺治疗组较之模型组神经元变性均明显减轻。模型组大鼠较正常组大鼠脑组织内Aβ蛋白显著升高。立即针刺组及针刺治疗组脑组织内Aβ蛋白显著下降。结论针刺能减轻神经元的变性受损,降低其脑组织内Aβ水平,减少Aβ的神经毒作用。这可能是针刺改善慢性脑缺血作用机制之一。Objective To study the acupuncture＇s impact on the changes in pathomorphism and Aβ protein level in the brain of rats with chronic cerebral ischemia and explore the intervention mechanism of acupuncture on the chronic cerebral ischemia.Methods This study established the chronic cerebral ischemic models by permanent occlusion of bilateral common carotid arteries（2VO）in rats,and observed the pathomorphologic changes the Aβ level changes in the rats＇ brain in four groups（the control group,the model group,the immediate acupuncture group,the acupuncture group）.Results The neuronal degenerative changes in rats＇ brain were more severe in the model group;while the degenerative changes were significantly improved in the immediate acupuncture group and the acupuncture group.The levels of Aβ in rats＇ brain were significantly elevated in the model group than those in the control group.In the immediate acupuncture group and the acupuncture group,the rats＇ Aβ levels were significantly reduced.Conclusion The acupuncture can improve the neuronal degenerative changes in rat＇s brain,and effectively lower the level of the Aβ in rat＇s brain as well as reduced toxicity to nerve.This is possibly one of the mechanisms that explain acupuncture can improve chronic cerebral ischemia.
