摘  要:目的:探讨低氧预适应(HP)对大鼠自体肝移植后肠道TLR4蛋白表达的影响。方法:将SD大鼠随机分为正常对照组(NC)、自体肝移植(AT)组和低氧预适应+AT(HP)组3组,每组24只,采用改良的自体肝移植模型。HP组术前用8%氮氧混合气体处理90 min,分别于术后1、6、12 h处死大鼠取小肠标本,抽血检测血清ALT及AST水平,采用免疫组化法测定各组小肠组织TLR4蛋白表达情况,及应用逆转录-聚合酶反应(RT-PCR)检测小肠组织TNF-αmRNA的含量。结果:HP组血清ALT、AST水平,小肠组织TLR4蛋白表达及TNF-αmRNA的含量均显著低于AT组,但均高于NC组,差异均有统计学意义(P<0.05)。结论:HP通过下调肝移植大鼠小肠组织TLR4蛋白的表达,进一步抑制TLR4信号转导通路,减少TNF-α等细胞因子的合成与释放,从而减轻对移植肝功能的损伤。Objective: To investigate the impact of hypoxia preconditioning on the expression of intestinal TLR4 protein after orthotopic liver autotransplantation in rats intestinal.Methods: A modified orthopotic liver autotransplantation model was used to investigate the ischemia reperfusion injury in liver transplantation.Sprague Dawley rats were randomly divided into the following three groups: normal control(NC)group,autotransplantation(AT)group,and hypoxia preconditioning(HP)group,there were 24 rats in each group.HP Group was given an 8% oxygen_mixed gas for 90 minutes before the operation.At 1,6 and 12 hours after operation,ras were killed and following tests were conducted: 1).blood was drawn to conduct a chemical examination,2)expression of TLR4 protein in intestinal was observed with immunohistochemical stains,3)use RT-PCR to observe the expression of TNF-α mRNA.Results: Serum levels of ALT,AST,and expression of TLR4 protein and TNF-α mRNA in intestinal of HP group were significantly lower than AT group(P0.05) at 1,6 and 12 hours after the orthopotic liver autotransplantation.Conclusion: HP can restrain TLR4 signaling pathway by down regulating the expression of TLR4 protein in orthotopic liver autotransplantation in rats,thus inducing the release of multi-kinds of cytokines like TNF-α and inflammatory mediators,and finally mitigating the damage of transplanted liver.
