摘 要:目的筛查低剂量长波紫外线(UVA)诱导培养人皮肤黑素细胞适应性反应的相关基因,探讨适应性反应的分子机制。方法取5—10代培养的人皮肤黑素细胞,实验组以7.2J/cm^2 UVA照射,每天1次,共4次,于最后1次照射6h后以致死量UVA(86.4J/cm^2)照射;对照组为具有相同传代数的黑素细胞,直接以86.4J/cm^2 UVA照射。用人类全基因组寡聚核苷酸芯片筛选黑素细胞差异表达基因,按照国际通用分类标准对结果进行功能分类,利用实时荧光定量PCR技术对筛选出来的部分差异表达基因进行分析验证。结果低剂量UVA诱导的培养人皮肤黑素细胞的适应性反应中,出现表达差异的基因129个,其中表达上调的基因有81个,表达下调基因48个,这些基因主要与代谢、原癌基因和抑癌基因、转运、信号转导、细胞凋亡以及DNA合成及修复有关;实时荧光定量PCR检测结果与芯片一致。结论培养人皮肤黑素细胞在低剂量UVA诱导的适应性反应中出现大量基因的差异表达,为研究低剂量UVA适应性反应的分子机制提供了重要基础。Objective To screen for genes associated with low-dose UVA irradiation:induced adaptation reaction in cultured human melanocytes, and to explore the molecular mechanism of the adaptation. Methods Cultured human melanoeytes of fifth to tenth passage were divided into two groups, experimental group was subjected to an irradiation with UVA at 7.2 J/cm2 once daily for 4 times and an additional irradiation at a lethal dose of 86.4 J/cm^2 6 hours after the above 4-session irradiation, and control group subjected to a single irradiation with UVA at 86.4 J/cm^2. A human genome-wide oligonucleotide chip was used to screen for differentially expressed genes between the two groups of cells followed by a functional classification based on international standard. Moreover, a part of these genes were analyzed and identified by real-time fluorescent quantitative PCR. Results In the adaptation reaction induced by low-dose UVA irradiation in cultured melanocytes, there were 129 differentially expressed genes, including 81 up-regulated genes and 48 down-regu- lated genes. These genes were found to be mainly involved in metabolism,transport, signal transduetion, apoptosis, DNA synthesis and repair, and some of them were oncogenes or anti-oncogenes. Real-time PCR confirmed some of the differentially expressed genes. Conclusions The whole genome-wide oligonucleotide chip could screen with high efficiency for differentially expressed genes in low-dose UVA irradiation-induced adaptation reaction in cultured melanocytes.
