摘  要:目的探讨布地奈德对哮喘气道重塑大鼠原代培养的气道平滑肌细胞(ASMCs)上清液中白细胞介素13(IL-13)及干扰素γ(IFN-γ)含量的影响。方法 60只Wistar大鼠随机分为三组,每组20只。A组吸入卵清蛋白制作哮喘气道重塑模型,B组在A组基础上隔日1次吸入布地奈德气雾剂,C组吸入生理盐水作对照。8周后取材行苏木素-伊红(HE)染色,测量各组大鼠肺内支气管气道壁面积(Wat)/气道基膜周长(Pbm)及平滑肌层面积(Was)/Pbm;原代培养大鼠ASMCs,ELISA法检测各组ASMCs上清液中IL-13及IFN-γ含量。结果 A、B组Wat/Pbm及Was/Pbm均较C组增加,但B组较A组降低(P<0.05)。A组IL-13含量较C组增加,IFN-γ含量较C组降低(P<0.05);B组IL-13含量较A组降低,IFN-γ含量较A组增加(P<0.05)。结论布地奈德通过降低哮喘大鼠ASMCs间质中IL-13及增加IFN-γ的表达来减轻气道重塑。Objective To investigate the effects of budesonide on the levels of interleukin 13(IL-13) and interferon gamma(IFN-γ) in the supernatant of airway smooth muscle cells(ASMCs) in asthmatic rat.Methods Sixty Wistar rats were randomly assigned into three groups with 20 rats each.The rats in group A were sensitized with ovalbumin aerosol for 8 weeks as asthmatic airway remodeling models and those in group B were given additional budesonide aerosol every other day for 8 weeks.The rats in group C were given saline inhalation as the controls.After 8 weeks the rat lungs were taken for HE staining and image analysis was used to measure wall area normalized by basal membrane perimeter(Wat/Pbm) and smooth muscle layers area normalized by basal membrane perimeter(Was/Pbm) in lobar bronchi.The levels of IL-13 and IFN-γ in the supernatant of ASMCs were detected by ELISA.Results Wat/Pbm and Was/Pbm in groups of A and B were higher than those in group C,while those in group B were lower than those in group A(P0.05).Compared with group C,the level of IL-13 increased,while IFN-γ decreased,significantly in group A(P0.05).Compared with group A,the level of IL-13 decreased,while IFN-γ increased,significantly in group B(P0.05).Conclusion Budesonide is able to decrease the level of IL-13 and increase the level of IFN-γ in ASMCs stroma to alleviate airway remodeling.
