摘 要:目的探讨补阳还五汤(BYHWD)对脊髓损伤(SCI)大鼠脊髓组织超微结构的影响,从微观角度探讨脊髓损伤的病理机制。方法应用SPF级Wistar大鼠,采用Allens’打击法制作中度SCI模型,参照改良Tarlov评分标准,分为模型组、BYHWD组、金纳多组,正常组不造模;用药2周结束实验后,取SCI大鼠脊髓组织制片行透射电子显微镜超微结构观察。结果正常组大鼠胞内与灰质内均未观察到空泡;模型组大鼠脊髓损伤严重,大部分的线粒体嵴丢失,形成空泡,内质网重度扩张,与其他组别比较有明显差异,差异有统计学意义(P<0.01);BYHWD组与模型组和金纳多组比较有明显好转,差异有统计学意义(P<0.01)。大鼠后肢运动功能Tarlov评分结果:在0 d~3周内,正常组大鼠后肢运动功能Tarlov评分无明显变化;BYHWD组与金纳多组评分随时间变化差异无统计学意义(P>0.05),与模型组及正常组比较差异有统计学意义(P<0.05)。结论 BYHWD能有效保护损伤脊髓组织的组织结构继发性损伤,营养神经元细胞,促进神经纤维的修复。Objective To explore the influence of Buyang Huanwu Decoction(BYHWD) on the ultrastructure in spinal cord tissue of the rats with spinal cord injury(SCI),to explore the pathological microscopic mechanism of SCI.Methods Wistar rats in SPF grade were used for the experimental animals.The moderate SCI models were built based on modified Allen's method.The model rats were randomly divided into model group,BYHWD group and Ginaton group.Normal control group was also set up.Rats were treated with BYHWD or Ginaton for 2 weeks.The modified Tarlov criteria was used for the evaluation of the hind limb motor function of rats.At the end of the treatment,spinal cord tissue slices were stained by hematoxylin and eosin(HE).Ultrastructure of the tissue was observed under transmission electron microscope(TEM).Results No vacuolus was found in the cytoplasm and grey matter of normal group.In the model group,spinal cord was badly hurt: most of mitochondrial cristae disappeared,vacuolus formed,endoplasmic reticulum were swollen badly.The differences of the above changes in the model group were significant as compared with other three group(P 0.01).Compared to the model group and Ginaton group,the ultrastructure of spinal cord in BYHWD group was improved,and the difference was statistical significance(P 0.01).The score of hind limb motor function evaluated by Tarlov criteria was as follows: Tarlov scores in the normal group were unchanged in 3 weeks,the scores in BYHWD group and Ginaton group changed with time,and the scores in BYHWD group showed no statistical significance compared with Ginaton group(P 0.05),but was statistical difference compared with model blank group and normal group(P 0.05).Conclusion BYHWD can effectively protect spinal cord from secondary lesion,nurture neurons and promote the recovery of nerve fibers.
