健脾益气清幽汤对慢性浅表性胃炎大鼠血清IL-2及胃组织IL-2mRNA的影响

摘　　要：目的探讨健脾益气清幽汤对慢性浅表性胃炎(CSG)大鼠血清白细胞介素-2(IL-2)及胃组织IL-2mRNA的影响。方法将66只Wistar大鼠随机分为空白对照组、模型组、健脾益气清幽汤低、中、高剂量组及兰索拉唑组,用0.02%氨水给大鼠自由饮用,连续90 d,制备大鼠试验性CSG模型。用HE染色检测动物造模效果,用RT-PCR检测胃组织IL-2mRNA表达,放射免疫法检测血清IL-2含量。结果健脾益气清幽汤各剂量组大鼠胃组织IL-2mRNA表达明显增多,与模型组比较有显著性差异(P<0.01);各剂量组大鼠血清IL-2含量明显升高,与模型组比较有显著性差异(P<0.05)。结论健脾益气清幽汤能明显提高胃组织IL-2mRNA表达及血清IL-2含量,这可能是健脾益气清幽汤有效治疗CSG的机制。Objective To investigate the effect of Jianpi Yiqi Qingyou Decoction（JYQD） on serum IL-2 content and gastric IL-2 mRNA in rats with chronic superficial gastritis（CSG）.Methods Wistar rats were randomly divided into 6 groups（11 rats in each group）： blank control group,model group,lansoprazole group,and low-,middle-and high-dose JYQD groups.The rat models of CSG were established by drinking 0.02% ammonia freely for continuous 90 days.Gastric pathological changes were detected by HE staining.Reverse transcriptase polymerase chain reaction（RT-PCR） was used for the detection of the expression of IL-2 mRNA in gastric tissue,and radioimmunoassay was used for the detection of the content of IL-2 in serum.Results Compared with those in the model group,the expression of IL-2 mRNA in gastric tissue（P 0.01） and the content of IL-2 in serum（P 0.05） obviously increased in JYQP groups.Conclusion JYQD can obviously improve the expression levels of IL-2 mRNA in gastric tissue and the content of IL-2 in serum,which may be one of the therapeutic mechanisms of JYQD for chronic superficial gastritis.
