摘 要:目的研究某市典型环境水有机提取物的细胞毒性及致突变性。方法采集某市饮用水源水、城市回用水、河水及公园景观水水样,以固相萃取技术提取水中有机物,以MTT法检测有机提取物对中国仓鼠肺细胞(CHL细胞)的细胞毒性,以Ames试验检测有机提取物的致突变性。结果水源水、城市回用水、河水、景观水有机提取物引起CHL细胞毒性的IC50分别为550、90、250、500 ml/ml。Ames试验显示,在无代谢活化系统下,城市回用水在500 ml/皿及以上、河水在1 000ml/皿及以上TA98和TA100两菌株均呈阳性;景观水在2 000 ml/皿TA98呈阳性;在有代谢活化系统下,城市回用水TA98菌株在500 ml/皿及以上、TA100在1 000 ml/皿及以上,河水在1 000 ml/皿及以上两菌株均呈阳性,水源水及景观水未见致突变性。结论城市典型环境水有机提取物在一定剂量下,对CHL细胞均产生毒性作用。在无代谢活化系统下,城市回用水、河水及公园景观水有机提取物具有致突变作用。在有代谢活化系统下,城市回用水、河水有机提取物具有致突变作用。Objective To study mutagenic effect and cytotoxicity of typical environmental water in a city. Methods Reverse phase C-18 solid-phase extraction (RP-C 18SPE) was used for the extraction of compounds from the water samples collected from reclaimed water, water source, river water and scenic water. MT]~ and Ames test were used to detect cytotoxicity and genetoxicity respectively. Results MTr test showed that all the four kinds of water could induce decrease of viability of CHL cells. Reclaimed water and river water could induce mutation of TA98 and TA 100 strains with or without S9 at the dose of 500 ml/dish or 1 000 ml/dish. Scenic water could induce mutation of TA98 and TA100 strains only with $9 at the dose of 1 000 ml/dish. Negative results were found in source water. Conclusion In a city, the organic extract of environmental water is cytotoxic at a certain exposure level. The organic extract of reclaimed water and river water is mutagenic before and after metabolic activation and that of scenic water may lose mutagenicity after metabolic activation.
