摘  要:目的:研究复方六月青(CLYQ)对肝星状细胞(HSC)I型胶原(Col I)合成的影响。方法:采用四甲基噻唑蓝(MTT)法检测CLYQ对细胞的毒性,分别采用免疫细胞化学法和实时荧光定量PCR法检测CLYQ对HSC的Col I蛋白和mRNA表达的影响。结果:CLYQ对HSC毒性很低。CLYQ各剂量组中HSC的Col I蛋白表达显著减弱(P<0.01)。CLYQ高、中剂量组均可显著下调HSC的Col I mRNA表达(P<0.01)。结论:CLYQ通过下调HSC的Col I蛋白及mRNA表达,从而减少细胞外基质(ECM)合成,发挥抗肝纤维化作用。Objective:To determine the effect of compound Liuyueqing(CLYQ) on the synthesis of collagen type I(Col I) in hepatic stellate cells(HSCs).Methods:The expression of Col I protein and mRNA in HSCs was detected by immunocytochemistry and real-time fluorescence quantitative PCR,respectively.Results:Col I protein expression in the HSCs was significantly weakened after CLYQ treatment at all concentrations(P〈0.01).The expression of Col I mRNA in HSCs was significantly down-regulated after CLYQ treatment at higher concentrations(P〈0.01).Conclusion:CLYQ can down-regulate the expression of Col I mRNA and protein in HSCs,result in reduction of extracellular matrix(ECM) synthesis,and may play a inhibitory role in hepatic fibrosis.
