摘  要:通过利用RAPD(Random Amplified Polymorphic DNAs)随机引物中筛选到的可扩增出清晰条带、主带明显、稳定的10条引物,对来自安徽合肥、淮南、和县、潜山、岳西、江苏南京和四川邛崃等县市的发病辣椒上分离鉴定获得23个辣椒疫霉菌(Phytophthora capsici)进行全基因组DNA RAPD标记遗传多样性分析。选用引物共标出DNA指纹图带113条,其中多态性条带96条,多态性检测率为84.96%,表明辣椒疫霉菌具有较丰富的遗传多样性。根据引物扩增的DNA指纹图谱,运用UPGMA法分析,以遗传相似系数0.7为阈值,供试菌株被划分为3个遗传聚类组,除部分相同地理来源的菌株被划分为同一组外,其他不同菌株间的遗传相似性与地理来源无直接相关性。Random amplified polymorphic DNA markers were used to analyse genetic diversity among the 23 isolates of Phytophthora Capsici obtained from the infected peppers collected in Hefei,Huainan,Hexian,Qianshan,Yuexi,Nanjing and Qionglai.10 random primers were selected.A total of 113 reproducible RAPD fragments were scored,of which 96(84.96%) were polymorphic.This showed that the polymorphism existed among different isolates.The genetic distance and phylogenetic tree were constructed by using of UPGMA cluster analysis.The analysis indicated that the isolates were classified into three groups according to the similarity coef-ficient of 0.7.Only a few isolates from the same locality were divided into the same group and there was no cor-relation between DNA polymorphism and their geographical origins.
