离子对色谱法测定颇得斯安灌肠剂中美沙拉嗪的含量及其降解产物

摘　　要：报道了用四丁基氢氧化铵离子对试剂,在C8色谱柱上同时测定美沙拉嗪及其降解产物2,5-二羟基苯甲酸的含量,流动相为乙睛-0.2mol／L枸橼酸钾缓冲液-0.3％的四丁基氢氧化铵溶液（1：5：80）,采用荧光检测器,λEX为315nm,λEM为430nm。美沙拉嗪线性范围为0.1～0.8mg／ml,相关系数为0.9999,平均回收率为99.4％（n=5）,RSD=0.2％;2,5-二羟基苯甲酸线性范围为0.26～2.0μg／ml,相关系数为0.9999,平均回收率为99.0％（n＝5）,RSD＝0.4％。A reversed phase ion-pair HPLC method was established for the determination of mesalazine and gentisic acid in Pentasa rectal suspension. Mesalazine and gentisic acid can be separated on Spherisorb octyl 5 μm column with a mobil phase of acetonitrile-0. 2 mol/L potassium citrate buffer-purified water (15: 5: 80, V/V/V) contalning 0. 3 % of tetrabuthyl ammonium hydroxide solution and FLD detector, XEX = 315nm, λEM=430 nm. The flow rate was 1 ml/min. The stundard curve of mesalazine was linear in the concentration range of 0.1-0. 8 mg/ml and the correlation coefficient was 0. 9999. The average recovery and relative standard deviation were 99.4% (n=5) and 0. 2 %,respectively. The standard curve of gentisic acid was linear in the concentration range of 0. 26-2.0 (g/ml) and the correlation coefficient was 0. 9999. The average recovery and relative standard deviation were 99. 0% (n=5) and 0.4%,respectively.
