摘 要:目的研究内皮素-1反义脱氧寡核苷酸(ET-1AS-ODN)对链脲佐菌素(streptozotocin,STZ)诱导的糖尿病性心肌病变大鼠心肌组织形态学的影响及其机制。方法一次性腹腔注射STZ(60mg/kg),建立大鼠糖尿病模型,观察5周,使之形成心肌损伤,并观察ET-1AS-ODN预防性给药后心肌组织形态学的变化。结果实验动物饲养5周后,与生理盐水对照组相比,糖尿病模型组大鼠一般状况差,体重明显降低(P<0.05),心肌纤维横纹模糊,局灶性心肌纤维溶解、变性、坏死;与DM组比较,ET-1AS-ODN各组大鼠精神状态有所改善,心肌纤维排列较整齐。结论内皮素-1反义寡核苷酸可减轻STZ诱导的糖尿病大鼠的心肌组织形态学损伤,对心肌组织具有保护作用,其机制与减少内源性内皮素的产生有关,表明内皮素-1在糖尿病心肌损伤中起重要作用。Objective To investigate the protective effects of endothelin-1 mRNA antisense oligodeoxynucleotide(ET-1AS-ODN) on the morphology of cardiomyopathy(DC) and to discuss the pathogenesis and mechanism of DC in a rat model.Methods Diabetic cardiomyopathy rats were developed for 5 weeks on the diabetic model induced by intraperitoneal administration of streptozotocin(STZ) at 60mg/kg and the morphological changes of cardiomyocytes were observed after the ET-1AS-ODN was administrated preventively.Results Compared with the normal controls,the general condition and body weight of the DM group decreased significantly(P0.01).Myocardial fiber saluted,degenerated and necroses barfly.Compared with the DM group,the general condition of ET-1AS-ODN groups was improved.The myocardial fiber arranged neatly.Conclusion Endothelin-1mRNA antisense oligodeoxynucleotide can prevent myocardium from injury in diabetic rats and has a protective effect on the myocardium.It indicates that ET-1 does play an important role in the pathogenesis and mechanism of cardiomyopathy in diabetes.
