HPLC法测定热炎清颗粒中虎杖苷和白藜芦醇的含量

摘　　要：目的:建立同时测定热炎清颗粒中虎杖苷和白藜芦醇含量的方法。方法:采用HPLC法,色谱条件为Luna C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈(A)-0.4%(B,V/V)冰醋酸溶液梯度洗脱(0-40min,A18%-57%,B82%-43%),流速:1.0 mL·min-1,柱温:25℃,检测波长:284 nm。结果:虎杖苷和白藜芦醇分别在0.031-0.62 mg·mL-1(r=0.9998)和0.007-0.130mg·mL-1(r=0.9998)浓度范围内有良好的线性关系,平均加样回收率分别为98.8%,98.3%;RSD%均小于2.0%。结论:本方法准确,重复性好,可用于该制剂的质量控制。Objective To establish a method for the determination of polydatin and resveratrol in Reyanqing Granules.Method The HPLC system consisting the Luna C18（4.6 mm×250 mm,5 μm）column,the mobile phase consisted of Acetonitrile（A）-0.4%（B,V/V）HAc with the gradient elution（0-40min,A18%-57%,B82%-43%）,the flow rate was 1.0 mL·min-1,the column temperature was 25 ℃,The UV detector was set at 284 nm.Results The linear response range was 0.031-0.62 mg·mL-1（r=0.9998） and 0.007-0.130 mg·mL-1（r=0.9998）,respectively.The average recovery of protocatechuic acid and protocatechualdehyde were 98.8% and 98.3%,respectively,RSD2.0%.Conclusions The assay demonstrated that the method had adequate and selectivily to measure the concentrations of accurate,repeatable,it can be applied in quantitative determination of polydatin and resveratrol in Reyanqing Granules.
