摘 要:目的观察解毒脱瘾汤(JTD)对吗啡依赖大鼠大脑内不同脑区多巴胺及其代谢产物的影响。方法采用大鼠吗啡依赖戒断模型,观察解毒脱瘾汤对吗啡依赖大鼠大脑内不同脑区多巴胺及其代谢产物的改变。结果成瘾组大鼠PFC和NAc的DA含量与对照组比较显著增高,差异有统计学意义(P<0.01),而纹状体的DA含量与对照组比较显著降低,海马的DA含量两组之间差异无统计学意义;解毒脱瘾汤中、高剂量组与可乐定组均可降低海马、PFC、NAc的DA含量,差异有统计学意义(P<0.01),但对纹状体的DA含量影响不大,差异无统计学意义;成瘾组大鼠PFC和NAc的DOPAC含量与对照组比较显著增高,成瘾组大鼠纹状体的DOPAC比对照组低,差异有统计学意义(P<0.01),而两组大鼠海马中均未能检测到DOPAC;解毒脱瘾汤中、高剂量组与可乐定组均可降低PFC、NAc的DOPAC含量,对纹状体的DOPAC含量影响不大,差异无统计学意义。结论吗啡依赖大鼠脑内可能存在多巴胺及其代谢产物障碍,解毒脱瘾汤可能通过改善吗啡依赖大鼠多巴胺物质含量变化,消除其戒断症状,从而达到治疗的目的。Objective To measure the effects of Jiedu Tuoyin decoction(JTD) on brain dopamine and its metabolites in morphinistic rats.Methods The detoxification models of morphinistic rats were used to observe the effects of JTD on the brain dopamine and its metabolites in different brain regions.Results Compared with the control group,the DA content of PFC and the NAc in addicted group were significantly higher(P〈0.01),while the DA content in striatum was lower,and the DA content in the hippocampus was not significant;in the middle-dose and high-dose of JTD groups and the clonidine group,the DA content of hippocampus,PFC and NAc lowered(P〈0.01);the DA content in the striatum was not so affected in the each dose of JTD groups and the clonidine group,which was no statistical significant difference;compared with the control group,the DOPAC content of PFC and NAc in addicted group were significantly higher,and DOPAC in striatum was lower(P〈0.01),while DOPAC were not detected in the hippocampus;all the DA contents of PFC and NAc in the middle-dose and high-dose of JTD groups and the clonidine group lowered,and the striatal DA content was not influenced in those groups,the statistics difference was no significant.Conclusion There may exists obstacles of brain dopamine and its metabolites in brain of morphinistic rats,and JTD may be effective to improve the morphine content of dopamine material changes for elimination of the withdrawal symptoms.
