连萸降压胶囊对自发性高血压大鼠血清蛋白质组学分析

摘　　要：目的:研究连萸降压胶囊对自发性高血压大鼠(Spontaneously hypertensive rat)血清蛋白质表达的影响。方法:选取SHR 20只,随机数法分2组,模型组予0.9%生理盐水灌胃;中药组予连萸降压胶囊灌胃,连续灌胃4周后,取主动脉血,分离血清,每组随机各取5份血清样品。用双向凝胶电泳等方法测定肽质量指纹图谱(PMF),鉴定并获取差异表达蛋白质的信息。结果:2组凝胶上的平均蛋白点数分别为476±30、504±45。连萸降压胶囊干预SHR后,有22个蛋白表达发生了显著变化,表达增强的15个,表达降低的4个,新增加蛋白点3个。22个差异蛋白点进行质谱分析,鉴定出的10个蛋白质为视黄醇结合蛋白、E3泛素连接酶、线粒体膜蛋白、锌指蛋白、T细胞受体、转甲状腺蛋白A-链、Rho GTPase、methyltransferase,UbiE/COQ5 family、2个假想蛋白。结论:连萸降压胶囊降压机制,可能与促使E3泛素连接酶等8种蛋白高表达,促使视黄醇结合蛋白、Rho GTPase低表达有关。Objective：To observe the effect of LY capsule on serum protein expression in SHR.Methods： 20 SHR were selected and randomly dividedinto two groups;The model group were treated by saline only while the Chinese medicine group were gavaged by suspension made by LY capsule.After 4 weeks successive gavaged,taking aortic blood and centrifugating.5 serum samples were took from each group to use two-dimensional gel electrophoresis,Database search identified the protein corresponding to the peptide and obtained the information of the different expression protein.Results： In two groups,the average protein points were 476 ± 30、504 ± 45.Compared with normal group,there were 22 significantly different points,4 reduced expression and 15 enhanced expression and 3 additional points in model group;22 protein points were identified by MS： Retinol-binding protein,E3 ubiquitin ligase,mitochondrial membrane protein,zinc and lipoproteins,RhoGTPase,T-cellreceptor,Chain-A,RatTransthyretin,methyltransferase,UbiE /COQ5 family,2-hypothetical-protein.Conclusion： The antihypertensive mechanism of LY capsule may be related to the high expression of 8 protein,such as the E3 ubiquitin ligase,and the low expression of 2 protein.
