绵羊痘病毒的分离及双抗夹心法ELISA的建立

摘  要:为了检查现有疫苗与山西北部舍饲羊群所患病是否匹配,以及更好地研究羊痘病毒和研制新型疫苗,对2009年山西北部绵羊群发生的疑似绵羊痘进行了病毒分离及ELISA检测研究。取疑似绵羊痘病羊的皮肤丘疹、水泡或脓泡组织的病毒悬液,接种2月龄绵羊睾丸细胞观察到明显的细胞病变。病毒悬液接种10日龄鸡胚绒毛尿囊膜,未出现痘斑。收获产生病变的细胞培养物,继而利用双抗夹心法ELISA检测羊痘病毒,并确定其病毒含量。结果表明,所分离的病毒为绵羊痘病毒,样品OD值平均为0.156,浓度为2.5 IU.L-1,接近于标准品(2号)。本研究为今后研制新型羊痘疫苗奠定了基础。In order to check for matching of existing vaccine and sicken flocks in northern Shanxi,and research sheep pox virus and the new vaccines,a field strain of sheep pox virus was isolated,and the biological characteristics of the isolated virus were studied by cell culture and ELISA detection.The obvious lesions of new born lamb spermary cell were observed after the inoculation of virus suspension.The pathological changes of chicken embryos were not observed after inoculation.This study harvested cell cultures,then used the double-antibody sand wich ELISA to detect sheep pox virus and determine the amount of the virus.The results above demonstrated that the isolated virus was sheep pox virus.The OD value(0.156) and concentration(2.5 IU·L^-1) we got were close to the standard product 2.This study lays a foundation for the future development of sheep pox vaccine.
