摘  要:目的研究南宁市散发性腹泻病人中,星状病毒的感染情况以及基因特征。方法 2007年2月~2008年3月,收集南宁市某医院门诊腹泻患者粪便标本346份。选择针对星状病毒核糖核酸聚合酶[Ribonucleic Acid(RNA)-dependent RNA Polymerase,RdRp]基因的两对半引物,应用半巢式聚合酶链反应(Heminested Polymerase Chain Reaction,HPCR)扩增目的片段,纯化后连接至克隆载体,测定核苷酸序列。应用ClusterW进行序列同源性比对,用MEGA3.1按照邻接法构建系统进化树。结果在检测的346份粪便标本中,HPCR检出6份阳性,经进化树分析证实它们均为人星状病毒,检出率为1.73%。6株病毒RdRp基因的氨基酸序列呈高度保守。结论南宁市散发性腹泻病例中存在星状病毒感染。Objectives To characterize the prevalence and gene sequences of astrovirus in patients with sporadic gastroenteritis in Nanning City.Methods A panel of 346 stool samples were collected from diarrheal patients attending outpatient clinics of a hospital in Nanning City between February 2007 and March 2008,and screened for astrovirus by heminested polymerase chain reaction(PCR) targeting RNAdependent RNA polymerase(RdRp).Positive PCR products were purified and then cloned into plasmids for DNA sequencing.Alignments of nucleotide and amino acid sequences were done using Cluster W,and phylogenetic trees were constructed using MEGA 3.1 by the neighbor-joining method.Results Among the 346 samples tested,6(1.73%) were positive by heminested PCR,all of which were confirmed as human astrovirus by phylogenetic analysis.The RdRp amino acid sequences of the 6 astrovirus strains were highly conserved.Conclusion The study result has confirmed the infection of astrovirus in sporadic gastroenteritis in Nanning.
