奥曲肽对人肝癌BEL-7402细胞生长调控的实验研究

摘　　要：目的观察生长抑素类似物奥曲肽(OCT)对人肝癌BEL—7402细胞生长调控的影响。方法培养人肝癌细胞株BEL—7402至对数生长期。MTT比色分析法测定OCT对BEL—7402细胞增殖的影响;流式细胞术分析OCT在10～6 mol/L时对BEL—7402细胞周期和凋亡的影响;HE染色观察10—6 mol/L OCT作用后BEL—7402细胞形态学改变。结果 1)OCT在一定浓度范围内呈剂量依赖性方式抑制肝癌BEL—7402细胞的生长;2)BEL—7402细胞经10—6 mol/L OCT作用后细胞周期发生明显改变。3)10—6 mol/L OCT作用后BEL—7402细胞形态学发生明显改变。结论 OCT在抑制肝癌细胞增殖,促进凋亡中起着重要的作用。Objective To evaluate the effects and mechanisms of somatostatin analogue（octreotide, OCT） on the regulation of the growth of human hepatocellular carcinoma BEL—7402 cell line.Methods By cultivating hepatocellular carcinoma BEL—7402 cell line toward period of logarithm vegetal. MTT colorimetric assay was performed to evaluate cell proliferation;Cell cycle distribution of BEL—7402 cell was measured by flow cytometric analysis;Morphological change of BEL—7402 cell after OCT administration was observed by HE staining.Results BEL—7402 cells were suppressed by OCT and the inhibitory effect of OCT was dose—dependent.OCT of 10—6mol/l was used to BEL—7402 cells which led to remarkably changes in the cell cycle.There were a series of typical morphological changes in the BEL—7402 cell after OCT administration.Conclusions OCT plays an important role in inhibiting the proliferation and inducing apoptosis of BEL—7402 cells.
