半边旗提取物5F对肝细胞癌BEL-7402生长抑制作用及增殖细胞核抗原表达的影响

摘　　要：目的研究半边旗提取物5F对肝细胞癌BEL-7402细胞生长的抑制作用。方法 MTT法检测5F对BEL-7402细胞的生长抑制作用;用Hoechst33258荧光染色法检测细胞凋亡;流式细胞仪检测5F对细胞周期的影响;免疫细胞化学法检测5F对增殖细胞核抗原(PCNA)表达的影响。结果 5F抑制BEL-7402细胞的生长,其效果与5F的浓度和作用时间相关,24、48和72h的IC50分别为60.33、21.32和11.22 mg/L;荧光双染色法显示经5F作用后细胞出现变形,染色质浓缩,产生凋亡小体;细胞被阻滞在G2/M,PCNA阳性表达率降低。结论 5F在体外能有效地抑制BEL-7402细胞的生长,其诱导凋亡作用可能与下调增殖细胞核抗原(PCNA)表达有关。【Objective】To investigate inhibitory effects of 5F from Pteris semiinnata L.on proliferation of hepatocellular carcinoma（HCC） BEL-7402 cells.【Methods】BEL-7402 cells growth inhibition mediated by 5F was detected by MTT assay.Hoechst33258 staining was used to observe apoptosis.Flow cytometer was used to determine the effect of 5F on the content of DNA in cells.PCNA expression was determined by immunocytochemical method.【Results】5F displayed growth inhibitory activity against BEL-7402 cells with IC50 values of 60.33,21.32 and 11.22 mg/L for 24,48,and 72 hours respectively.5F induced apoptosis.BEL-7402 cells treated by 5F were arrested at G2/M.5F could depress PCNA expression level in BEL-7402 cells.【Conclusions】In vitro 5F inhibited significantly the proliferation of BEL-7402 cells and the inducing apoptosis,the mechanism may be related with down-regulation of PCNA expression.
