摘  要:【目的】对普通小麦(Triticum aestivum L.)品种7182与华山新麦草(Psathyrostac hyshuashanica)杂交后代中选育的矮秆种质B62进行农艺性状考察和外源遗传物质检测。【方法】以普通小麦-华山新麦草矮秆种质B62为材料,在田间农艺性状考察的基础上,采用细胞学及基因组原位杂交(根尖与花粉母细胞中期Ⅰ)技术,检测B62中的华山新麦草遗传物质。【结果】根尖细胞学鉴定表明,B62染色体条数为44条;根尖细胞染色体原位杂交(GISH)表明,B62携带2条华山新麦草染色体;花粉母细胞减数分裂Ⅰ中期染色体GISH结果显示,2条外源染色体能够配对,说明2条异源染色体是华山新麦草的1对同源染色体。【结论】矮秆种质B62为普通小麦-华山新麦草二体异附加系,其农艺性状优于其小麦亲本7182。[Objective] The study was to observe the agronomy and alien genetic chromatin of a dwarf germplasm B62 obtained by screening the offspring of hybridizing between Triticum aestivum L.cv.7182 and Psathyrostachys huashanica.[Method] Cytology analysis,genomic in situ hybridization(GISH) analysis on root tip and PMC Ⅰ of B62 were used.[Result] 44 chromosomes were observed after identificating root tip in cytology.With P.huashanica Ns genomic DNA as a probe,root tip genomic in situ hybridization(GISH) analysis identified two material carries P.huashanica chromosomes.Then it was concluded that B62 was a wheat-P.huashanica disomic or double monosomic addition line;and the addition line for pollen mother cell mid-meiosisⅠgenomic in situ hybridization(GISH).The results show that the two alien chromosomes can pair and arrange in equatorial plate.It shows that the two added chromosomes are a pair of homologous chromosomes in P.huashanica.[Conclusion] B62 is a dwarf material of the wheat-P.huashanica hybridization disomic addition line,which agronomic are traits better than the wheat parent-7182.
