摘 要:目的探讨时辰对罗哌卡因、氯普鲁卡因、利多卡因惊厥半数有效量(ED50)和半数致死量(LD50)的影响。方法 270只小鼠随机分为ED50和LD50两组,每组135只;每组再分为3个亚组,即罗哌卡因亚组、氯普鲁卡因亚组、利多卡因亚组,每亚组45只;按不同时辰每亚组再随机分为3个次亚组,每次亚组15只,分别于8:00、14:00和20:00腹腔注射罗哌卡因、氯普鲁卡因或利多卡因,以小鼠惊厥和死亡为观察指标,用序贯法测定各时间点的ED50及LD50。结果利多卡因在14:00的惊厥ED50显著低于8:00和20:00(P值分别<0.05、0.01),在14:00的LD50显著低于8:00(P<0.05),其余各时辰间ED50、LD50的差异均无统计学意义(P值均>0.05)。罗哌卡因在8:00的惊厥ED50和LD50均显著低于14:00和20:00(P值分别<0.05、0.01),其余各时辰间ED50、LD50的差异均无统计学意义(P值均>0.05)。氯普鲁卡因在8:00的ED50显著低于14:00(P<0.05),其余各时辰间ED50、LD50的差异均无统计学意义(P值均>0.05)。结论罗哌卡因、氯普鲁卡因和利多卡因的惊厥ED50均具有时辰依赖性。利多卡因、罗哌卡因的LD50具有时辰依赖性,氯普鲁卡因的LD50未发现时辰依赖性。Objective To observe the effects of chrono on the median lethal dose(LD50)and the median effective dose(ED50)of convulsion induced by anaesthetics(Ropivacaine,Chloroprocaine,and Lidocaine).Methods A total of 270 mice were evenly randomized into 2 groups for investigation of LD50 and ED50;the two groups were then further assigned to 3 subgroups,with each subgroup including 45 mice;and each subgroup was also evenly divided into 3 tertiary groups.Ropivacaine,Chloroprocaine and Lidocaine were intraperitoneally injected at 8:00,14:00 and 20:00,respectively.The convulsion and death of animals were observed.And the ED50 and LD50 values were determined by up-down sequential allocation at different time points.Results The convulsive ED50 of Lidocaine at 14:00 was significantly lower than those at 8:00 and 20:00(P0.05,0.01,respectively).The LD50 value of Lidocaine at 14:00 was significantly lower than that at 8:00(P0.05).The convulsive ED50 and LD50 values of Ropivacaine at 8:00 were significantly lower than those at 14:00 and 20:00(P0.05,0.01,respectively).The convulsive ED50 value of Chloroprocaine at 8:00 was lower than that at 14:00(P0.05).Chrono showed no significant effect on the LD50 values of Chloroprocaine.Conclusion The effects of convulsive ED50 values of Ropivacaine,Chloroprocaine and Lidocaine are chrono-dependent.The effects of LD50 values of Ropivacaine and Lidocaine also are chrono-dependent.The effects of LD50value of Chloroprocaine is not chrono-dependent.
