氯索洛芬贴剂缓解慢性肌肉痛觉过敏大鼠疼痛的作用

摘  要:目的:评价氯索洛芬贴剂在慢性肌肉痛觉过敏大鼠模型中的镇痛作用。方法:雄性SD大鼠32只,采用腓肠肌注射pH4.0的0.9%盐水建立肌肉痛觉过敏大鼠模型,根据给药的种类和时间不同将大鼠随机分为4组(n=8):造模前氯索洛芬组(BL组)、造模前薄荷组(BB组)、造模后氯索洛芬组(AL组)和造模后薄荷组(AB组)。BL组和BB组在腓肠肌首次注射pH4.0的0.9%盐水后第5d分别每天给予氯索洛芬贴剂和不含氯索洛芬的薄荷贴剂;AL组和AB组在腓肠肌首次注射pH4.0的0.9%盐水后第14d分别每天给予氯索洛芬贴剂和不含氯索洛芬的薄荷贴剂。所有大鼠首次注射pH4.0的0.9%盐水前1d(T0)、第1、5、6、8、10、12、14、16、19、20d时(T1~T10)采用von Frey纤维丝法测定大鼠的机械痛阈。结果:分别与BB组T5时相比,BB组大鼠的机械痛阈于T14时开始降低(P<0.05);分别与AB组T5时相比,AB组大鼠的机械痛阈于T6时开始降低(P<0.05);分别与各自的T5时相比,BL组大鼠的各时间点机械痛阈无改变,AL组大鼠的机械痛阈于T10时开始降低(P<0.05)。结论:氯索洛芬贴剂能减轻慢性肌肉痛觉过敏大鼠机械痛敏。Objective:To evaluate the effect of Loxoprofen on muscle hyperalgesia of chronic muscle pain in rats.Methods: Thirty-two male SD rats were divided into four groups randomly: loxoprofen-administration before model-completed,base-administration before model-completed,loxoprofen-administration after model-completed and base-administration after model-completed.The mechanical withdrawal thresholds were measured with von Frey filaments before injection and the 1st,5th,6th,8th,10th,12th,14th and 16th day after injection.Results:A significant decrease in the withdrawal threshold in the base-administration was observed compared with loxoprofen-administration on the 5th day.There were decreases in the withdrawal threshold both in the base-administration and loxoprofen-administration on the 14th day.A significant decrease in the mechanical withdrawal threshold in the loxoprofen-administration on the 14th day was also observed by test compared with loxoprofen-administration on the 5th day.Conclusion:Loxoprofen can reduce muscle hyperalgesia in model of chronic muscle pain in rats.
