摘  要:目的:研究大麻素CB1受体是否参与电针镇痛。方法:以完全弗氏佐剂造成AA大鼠模型,在电针治疗四次后以大麻素CB1受体拮抗剂AM251进行干预,检测各组大鼠缩爪反射潜伏期(PWL)的变化,并应用实时荧光定量PCR(FQ-PCR)和Western blot检测伏膈核、尾状核内CB1受体mRNA和蛋白质表达情况。结果:(1)电针治疗后大鼠PWL延长,电针+AM251组的电针镇痛效果显著弱于电针组(P<0.01);(2)电针+AM251组较电针组大鼠伏膈核、尾状核内CB1受体mRNA表达水平显著降低(P<0.01),但与假模型组和模型组比较,无显著性差异(P>0.05)。(3)Western blot结果与FQ-PCR结果一致:电针+AM251组较电针组大鼠伏膈核、尾状核内CB1受体蛋白质表达水平显著降低(P<0.01)。但与假模型组和模型组比较,无显著性差异(P>0.05)。结论:AM251能够翻转电针后大鼠伏膈核、尾状核内CB1受体的高表达,减弱电针镇痛作用,提示CB1受体可能参与电针镇痛作用。Objective: To explore the relationship between cannabinoid CB1 receptor and repeated electro-acupuncture(EA) analgesia.Methods: In the complete Freund's adjuvant-induced arthritis rats,paw withdrawing latency was investigated after repeated EA or the injection of cannabinoid CB1 receptor selective antagonist AM251.And the CB1 receptor mRNA was measured by using Real time fluorescence quantitative-polymerase chain reaction(FQ-PCR).The Western blot was used to observe the changes of CB1 receptor protein expression.Results:(1) The paw withdrawing latency was prolonged after the repeated EA while the analgesic effects of EA were attenuated after the injection of AM251(P〈0.01).(2) In the nucleus accumbens and caudate nucleus,the expression of CB1 receptor mRNA of EA+AM251 group was significantly decreased than that of repeated EA group(P〈0.01).There was no significant difference of gene expression of CB1 receptor between EA+AM251 group and sham model group or model group.(3)Western blot demonstrated the similar result as FQ-PCR: the CB1 receptor protein expression of EA+AM251 group was notably decreased than that of repeated EA group(P〈0.01). There was no significant difference of protein expression of CB1 receptor between EA+AM251 group and sham model group or model group.Conclusion: AM251 can attenuate the analgesic effects of repeated EA by reversing CB1 receptor higher expression,suggesting that CB1 receptor may play an important role in the EA analgesia.
