金欣口服液对呼吸道合胞病毒肺炎大鼠模型TNF-α、分泌型IgA的影响

摘　　要：目的:探讨中药复方金欣口服液对呼吸道合胞病毒肺炎模型幼年大鼠血清中TNF-α、支气管肺泡及小肠灌洗液中SIgA的影响。方法:将56只幼年大鼠随机分为正常组,模型组,金欣高、中、低剂量组及预先给药组,西药利巴韦林组,共7组。RSV滴鼻3天后灌胃给药,连续用药7天后处死。ELISA法测定各组大鼠血清中TNF-α、支气管肺泡及小肠灌洗液中SIgA的水平。结果:模型各组血清TNF-a及黏膜SIgA的含量表达水平均高于正常组(P<0.01)。与模型组相比,金欣高剂量、中剂量组血清TNF-a明显增高(均P<0.01),差异具有统计学意义,且金欣高剂量组与利巴韦林组比较有显著性差异(P<0.01);金欣预先给药组、高剂量组的支气管肺泡及小肠灌洗液和中剂量组支气管肺泡灌洗液中SIgA的表达均增高,差异具有统计学意义。其中预先给药组与高剂量组的支气管肺泡及高剂量组的小肠灌洗液和中剂量组支气管肺泡灌洗液与利巴韦林组比较有显著性差异(分别为P<0.01、P<0.05)。结论:金欣口服液可增加RSV肺炎模型幼年大鼠TNF-α、SIgA的分泌,并且存在一定的量效关系,是治疗RSV感染的有效方剂。Objective：To investigate the effects of Jinxin oral liquid on the secretion of the TNF-α in serum and SIgA in bronchoalveolar lavage fluid and intestinal fluid of the pneumonia rats induced by Respiratory Syncytial Viruses.Methods：56 Rats were randomly divided into seven groups,the normal control group,the model control group,low-dosage,medium-dosage and hig-dosage of Jinxin oral liquid groups,pretreated group and Ribavirin treated group.Rats in model control group,low-dosage,medium-dosage and hig-dosage of Jinxin oral liquid groups and Ribavirin treated group were treated with RSV use nasal drip method for three days,and then all the groups except the model control group were treated with Jinxin oral liquid or Ribavirin for seven days.At the end of the experiment,the rats were killed,and the blood samples and broncho alveolus lavage fluid and intestinal fluid were collected.The TNF-α in serum and SIgA in bronchoalveolar lavage fluid and intestinal fluid were measured by ELISA.Results：Compared with normal control group,the serum level of TNF-αin model control group was higer（P0.01）.The serum levels of TNF-αin the high and medium dosage of Jinxin oral liquid groups were increased,when compared with the model control group,moreover,the serum lelve of TNF-αin high dosage of Jinxin oral liquid group was increased more that compared with the Ribavirin treated group（P0.01）.The lelves of SIgA in bronchoalveolar lavage fluid and intestinal fluid were increased in Jinxin oral liquid pretreated group and high dosage of Jinxin oral liquid group,besides,the lelve of SIgA in bronchoalveolar lavage fluid was significantly increased.Conclusion：Jinxin oral liquid can increase the lelves of TNF-αand SIgA,furthermore there is a certain dose-effect relationship bewteen them.All in all,Jinxin orad liquid is an effective compound in treating infectous disease,such as the pneumonia induced by Respiratory Syncytial Viruses.
