摘 要:采用响应面法优化阴离子交换层析去除重组类人胶原蛋白(HLC)溶液中内毒素的工艺。利用单因素试验考察HLC质量浓度、NaCl浓度和pH值对内毒素去除率的影响,以内毒素去除率和HLC回收率为指标,对三因素进行中心复合设计,并经响应面法优化,分析得到二次多项式回归方程的预测模型。去除内毒素的最佳工艺条件为:HLC质量浓度0.95 mg/mL,pH值7.6,NaCl浓度50 mmol/L。在该条件下,内毒素的去除率及HLC回收率分别可达到97.92%和98.01%。Response surface analysis methodology(RSM) was used for optimizing the removal process of endotoxin from human-like collagen(HLC) solution based on anion-exchange chromatography.The effects of three crucial factors(HLC mass concentration,pH value and NaCl concentration) on endotoxin removal efficiency(ERE) and HLC recovery rate were investigated by using one-factor-at-a-time design.The response surface central composition design,in which the interactions between the variables were considered,was applied to optimize these factors.A quadratic polynomial regression model was developed.The optimal endotoxin removal efficiency conditions are as follows: the mass concentration of HLC is 0.95 mg/mL,pH=7.6 and NaCl concentration is 50 mmol/L.Under such conditions,the endotoxin removal efficiency and HLC recovery rate are 97.92% and 98.01% respectively.
