摘 要:目的探讨脑力苏胶囊对小鼠海马区脑红蛋白(neuroglobin,NGB)mRNA表达的影响。方法采用双侧颈总动脉结扎反复脑缺血再灌注的方法制备血管性痴呆小鼠模型。模型成功建立后,随机分为假手术、模型、喜得镇组、脑力苏胶囊高、低剂量组,其中后3组为治疗组。术后15天进行行为学实验,并采用反转录-聚合酶链反应(RT-PCR)检测实验小鼠海马组织NGB mRNA表达的影响。结果模型组小鼠学习成绩与记忆成绩下降,各治疗组小鼠学习成绩与记忆成绩均有提高。模型组小鼠脑组织海马区NGB mRNA的表达较假手术组增加,备用药组均较模型组增加。结论脑力苏胶囊对血管性痴呆小鼠有治疗作用,其作用机制可能与调节海马组织NGB mRNA的表达,减轻缺血再灌注损伤有关。Objective To investigate the influence of Nao in the hippocampus of mice red protein (neuroglobin, NGB) mRNA expression. Methods The bilateral common carotid artery ligation and repeated cerebral isehemia and reperfusion was prepared mouse model of vascular dementia. Model suceesstully established, were randomly divided into sham, model, Hydergine group Nao high and low dose group, where the latter three groups tbr the treatment group. 15 days after the be- havioral experiments, using reverse transcription - polymerase chain reaction (RT - PCR) detection of the hippoeampus of mice the expression of NGB mRNA. Results The model group decreased academic performance and memotT pertbrmanee, the treated mice were improved academic pertormance and memory. Model of hippocampal tissue in mice the expression of NGB mRN A increased cmnpared with sham group, the treatment group compared with model group increased. Conclusion Nao on vascular dementia has therapeutic eftect in mice and its mechanism may be related to regulating the expression of NGB mRNA in hippocampus and reduce ischemia-reperfusion injury.
