摘 要:目的:探讨静态负荷对大鼠骨骼肌损伤标志酶及肌细胞钙水平的影响。方法:96只健康雄性Wistar大鼠,随机分为12组,每组8只。将大鼠麻醉后游离左下肢比目鱼肌远端,分别施加0、25、50和100 g负荷,各负荷组持续时间分别为30、60和90 min,负荷结束后经腹主动脉取血,分离血清测定肌酸激酶(creatine kinase,CK)、乳酸脱氢酶(lactatedehydrogenase,LDH)。在4℃条件下,9 000 r/min离心20 min分离比目鱼肌胞浆液与线粒体,测定钙浓度。结果:与对照组比较,血清CK活力,随负荷增加(50 g负荷)和时间延长(60 min、90 min),血清CK活力升高(P<0.01和P<0.05)。血清LDH活力,在50g组负荷90 min,100 g组负荷30 min、60 min、90 min LDH活力升高(P<0.01和P<0.05)。骨骼肌胞浆钙水平,在50 g、100 g组负荷30 min、60 min、90 min钙水平升高(P<0.01和P<0.05)。大鼠骨骼肌线粒体钙浓度在负荷25 g组负荷90 min,线粒体钙浓度增加(P<0.05);50和100 g组负荷60 min、90 min,线粒体钙浓度增加(P<0.05)。结论:长时间高强度静态负荷作用于骨骼肌,可导致血清CK、LDH活力升高,骨骼肌胞浆钙和线粒体钙浓度增高。Objective:To discuss the effect of static load on marker enzyme of skeletal muscle injury and calcium of skeletal muscle in rats. Methods: 96 healthy male wistar rats were randomly divided into 12 groups(8 rats in every group).Distal end of soleus of lower left limb was isolated after anesthesia.Load at 0,25, 50,100g and time at 30,60,90min were given to different groups.Serum creatine kinase(CK) and lactate dehydrogenase(LDH) were measured.Calcium concentration in cytoplasm and mitochondria of soleus under the condition of 4 ℃,9 000r/min offcenter for 20 min were detected.Results:Compared with control,activities of serum CK increased significantly with load increasing(50g) and time prolonging(30,60min)(P〈0.01,P〈0.05).Activities of serum LDH increased significantly in the group of 50g load after 90min and the group of 100g load after 30,60,90min(P〈0.01,P〈0.05).Cytoplasma calcium increased significantly in the group of 50g load and 100g load after 30,60,90min(P〈0.01,P〈0.05).Mitochondrial calcium increased in the group of 25g load after 90min,50g and 100g loads after 60,90min(P〈0.05).Conclusion:Long time and high strength load on the skeletal muscle will create the increase of serum CK and LDH activities,and the increase of calcium in cytoplasm and mitochondria of skeletal muscle.
