摘  要:以辣椒品种"哈椒一号"为试材,以无菌苗子叶、下胚轴和带柄子叶为外植体,采用正交设计探讨不同外植体、激素组合和AgNO3浓度对不定芽分化的影响。结果表明:最适不定芽培养基为MS+6-BA 6.0 mg/L+IAA 2.0 mg/L+AgNO34.0 mg/L,pH 5.8,最高不定芽分化率达88.2%。6-BA与IAA组合使叶片不定芽分化频率明显提高,6-BA 6.0 mg/L+IAA 2.0 mg/L的组合能够达到最高的诱导率。AgNO3显著地提高了不定芽诱导的频率,且缩短了不定芽的发生时间。3种外植体的不定芽诱导率以带柄子叶最高,其次是子叶,下胚轴最差。Taking pepper varieties 'No.1 Hajiao'as test material,using cotyledons,hypocotyls and cotyledons as explants,petiole,the effect of different explants and hormone combination and concentration of volatile AgNO3 on bud differentiation were studied by the method of orthogonal design.The results showed that the optimum medium for shoots induction medium was MS+ 6-BA 6.0 mg/L+ IAA 2.0 mg/L+AgNO3 4.0 mg/L,pH 5.8,which made the shoot buds differentiation frequency of 88.2%.On the shoots differentiation period,the combination of BA and IAA could raise the differentiation frequency obviously.The 6-BA 6.0 mg/L +IAA 2.0 mg/L could give the highest response.The AgNO3 significantly increased the frequency and shortened the time for shoot regeneration.In addition,among of three explants(the cotyledons,hypocotyls and cotyledons with petioles),the frequency of shoot differentiation of cotyledon with petiole explants was highest,and then the cotyledon explants were less higher,the hypocotyl explants were the lower.
