摘 要:目的 了解真性红细胞增多症(PV)患者外周血T细胞受体(TCR)Vβ24 个亚家族基因的表达及其克隆特点。方法 采用逆转录多聚酶链反应(RTPCR) 检测3 例PV 患者外周血单个核细胞TCRVβ24 个亚家族基因表达情况;采用基因扫描分析T细胞的克隆特点。结果 3 例PV患者外周血仅表达4 ~14 个Vβ亚家族,主要为Vβ2 、Vβ3 、Vβ16 、Vβ21 和Vβ23 ;2 例患者表达的Vβ3 和Vβ23T细胞为克隆性增殖。结论 PV 患者外周血TCRVβ亚家族的优势表达和克隆性增殖可能是患者体内对抗恶性细胞的免疫反应特征。Objective To investigate the expression and clonality of TCR Vβ subfamily genes in the T cells in polycythemia vera. Methods The 24 genes of the TCR Vβ subfamily were amplified with peripheral blood mononuclear cells from 3 cases of polycythemia vera(PV) by RT PCR, then the PCR products were further analyzed by genescan technique to evaluating the clonality of the detectable TCR Vβ T cells. Results Only 4~14 of the 24 Vβ subfamily T cells were found in the 3 PV cases, the commonly expressed Vβ subfamily genes were Vβ 2, Vβ 3, Vβ 16 , Vβ 21 and Vβ 23 . Two clonal T cells of Vβ 3 and Vβ 23 were identified in 2 cases. Conclusions The skew distribution and clonal expansion of TCR Vβ subfamily T cells in patients with polycythemia vera might be a feature of the immune response for malignant cells.
