摘 要:目的:HSV 可引起多种疾病,阿昔洛韦(ACV) 治疗HSV 感染效果虽好,但易出现耐药毒株;新药Cidofovir 对ACV 耐药毒株效果好,毒性也大。HSV 疫苗临床实验效果也不好。为了解决这些问题,必须继续开发新药。虎杖是一种抗病毒范围广的中药,为了了解虎杖的抗病毒性能并开发利用,我们对虎杖乙酸乙酯萃取部分的抗HSV 药效进行了研究。方法:我们以ACV 为阳性对照,利用病毒细胞病变抑制及空斑减数实验观察了药效,并且用中效作用原理对药效进行了分析。结果:在HEp2 细胞系统中,ACV 对HSV1F 株直接杀灭、增殖的抑制及感染阻断的ED50 分别为13 .22 ,9 .09 和8 .71μg/ml;晶Ⅳ的相应数值分别为0 .09 ,0 .07 和0 .03μg/ml;晶Ⅳ的三个治疗指数(TI)分别为ACVTI的307 ,272 和609 倍。ACV 对HSV2 333 株直接杀灭、增殖抑制及感染阻断的ED50 分别为16 .45 ,10 .85 和18 .62μg/ml;晶Ⅳ的相应数值分别为0 .07 ,0 .09 和2 .29μg/ml;晶Ⅳ的三个TI分别为ACVTI的498 ,251 和17 倍。结论:晶Ⅳ抗HSV 药效比ACV 强,毒?OBJECTIVE:Herpes simplex virus can induce many diseases,although therapeutical effect of ACV on HSV infection is good,but resistant strains against ACV appear easily,and new drug cidofovir can cure the infection of ACV resistant strains,but it has obvious toxicity,the results of clinical trials of HSV vaccines are not good.In order to resoluve these problems,it is necessary to exploit new antiherpes drugs contineously.Polygonum cuspidati is a Chinese medicinal herb with wide antiviral range,understanding and exploiting it′s antiviral property,we observed antiherpes effects of ethylacetate extract component of it.METHODS:With ACV as positive control,we investigated antiherpes effects of ethyl acetate extract component,i.e. crystalline Ⅲ and crystalline Ⅳ,by way of CPE inhibition and plaque reduction assays,and analyzed effects with median effect principle.RESULTS:In HEp 2 cell system:the ED 50 s of direct virucidal activity,the inhibition effects on the multiplication and infection of HSV 1 F strain of ACV are 13.22μg/ml,9.09μg/ml and 8.71μg/ml respectively,the corresponding figures of crystalline Ⅳ are 0.09,0.07 and 0.03μg/ml respectively.The treatment indices of crystalline Ⅳ are 307,272,609 times of ACV TIs.The ED 50 s of diredct virucidal activity,the inhibition effects on the multiplication and infection of HSV 2 333 strain are 16.45μg/ml,10.85μg/ml and 18.62μg/ml respectively,the corresponding figures of crystalline Ⅳ are 0.07μg/ml,0.09μg/ml and 2.29μg/ml respectively.The treatment indices of crystalline Ⅳ are 498,251,17 times of ACV TIs.CONCLUSION:The antiherpes effects of crystalline Ⅳ is stronger than ACV,and the toxicity of crystalline Ⅳ is less than ACV,it should be worth exploitation.
