摘 要:目的考察氮酮、月桂醇、油酸等几种常用促渗剂在不同浓度和联合使用对氯诺昔康体外经皮渗透的影响。方法用改良的Franz扩散池作为离体透皮试验装置,用pH8的磷酸盐缓冲液作为接收液,以稳态流量(Js)、增渗倍数(ER)及滞后时间为考察指标,观察不同促渗剂对氯诺昔康经皮吸收的促进作用。结果油溶性促渗剂促渗效果要好于水溶性促渗剂。以丙二醇为溶剂时,油溶性促渗剂氮酮、薄荷醇、十二醇、油酸对氯诺昔康都有促进作用。其中,氮酮渗透促进作用最为显著,月桂醇次之,两者联合应用效果显著。结论氮酮及其联合促渗剂的应用对氯诺昔康经皮给药均有理想渗透促进作用。Objective To study the effect of such penetration enhancers as azone,dodecanol,and oleic acid on the transdermal permeability of lornoxicam in vitro in different concentrations and different ways.Methods Experiments were carried out using improved Franz diffusion cell with PBS(pH8) as the receptor solution.The steady permeation rate(Js),enhancing rate(ER) and the lag time were computed to evaluate the penetration effect.Results Oil-soluble penetration enhancers were more effective than water-soluble penetration enhancers.The oil-soluble penetration enhancers and PG were synergistic.The combination of different penetration enhancers could produce different effect.The permeation enhancing effect of azone was the most significant,followed by dodecanol,and the effect of combination of the two was remarkable.Conclusion Azone and application of joint penetration enhancers are ideal penetration enhancers for the delivery of lornoxicam across the skin.
