摘 要:目的:评价人参皂甙Rg3对人恶性黑素瘤细胞系WM451生长和凋亡及对化疗药物氮烯咪胺敏感性的影响。方法:采用电镜和TUNEL染色法观察人参皂甙Rg3对WM451细胞凋亡的影响,用MTT法检测细胞增殖及对化疗药物氮烯咪胺的敏感性。结果:人参皂甙Rg3对恶性黑素瘤细胞生长有抑制作用且与浓度呈正相关,人参皂甙Rg3处理组细胞凋亡率(84%)较对照组(3%)明显增加。MTT法显示人参皂甙Rg3可增强氮烯咪胺对WM451细胞的杀伤作用,IC50值由(170.66±18.88)μg/mL降至(84.79±8.78)μg/mL。结论:人参皂甙Rg3可增加WM451细胞对化疗药物氮烯咪胺的敏感性。Objective: To assess the impact of Gensenoside Rg3 on growth and apoptosis, and sensitivity of human malignant melanoma cell lines WM451 to dacarbazine. Methods: The apoptosis of WM451 cells was examined by electron microscopy and TUNEL staining method. The growth curve of WM451 ceils and their sensitivities to dacarbazine were determined by MTr assay. Results: Gensenoside Rg3 significantly inhibited the proliferation of WM451 cells in a dose - dependent manner. In the group treated with Gensenoside Rg3, the apoptosis rate was 84%, which was higher than that in control group (3%). MTT assay showed Gensenoside Rg3 enhanced the anticancer toxicity of dacarbazine to WM451 cells, and the IC50 value was decreased from 170.66 ±18.88μg/mL to 84.79± 8.78 μg/mL. Conclusion: Gensenoside Rg3 can enhance the sensitivity of WM451 cells to dacarbazine.
