摘  要:应用"311-B"回归最优设计研究毒莠定等3种试剂对郁金香胚性愈伤组织诱导及胚状体产生的影响。结果表明:胚性愈伤组织出愈率及胚状体产生与毒莠定浓度呈极显著相关,毒莠定对郁金香胚性愈伤组织的诱导起决定性作用,培养20d愈伤组织就可以产生,最佳培养基配方为MS培养基+毒莠定6mg/L+BA0.5mg/L,出愈率达100%,每块愈伤组织平均产生10个胚状体。毒莠定与BA呈正交互作用,可以配合使用;与恶霉灵呈负相关,不必配合使用。The effects of 3 reagents such as picloram on tulip callus induction and somatic embryogenesis were studied by using the optimal design of"311-B"regression.The results showed that the callus inductivity and the somatic embryogenesis had a very significant correlation with picloram concentration.Picloram played a decisive role in tulip callus induction,and the callus could occur through 20 d culture.The optimal medium was MS+ 6 mg/L picloram+ 0.5 mg/L BA,the callus inductivity reached 100%and each piece of the callus averagely produced 10 embryoids.Picloram and BA had a positive interaction and could be used together.But picloram had a negative interaction with hymexazol and both did not have to be used together.
