摘  要:目的:比较测定几种不同动物骨骼肌肌球蛋白重链(MHC)亚型及成肌调节因子(MRFs)基因表达,引物均采用大鼠基因号设计,旨在比较观察其它动物与大鼠基因表达的交叉及相似度。方法:选用大鼠、家兔、牛与人的股外侧肌样本各6个,迅速用TRIZOL试剂匀浆固定,采用实时荧光-PCR法,测定骨骼肌MHCI,IIa,IIx,IIb四种亚型及成肌调节因子(MRFs)的mRNA基因表达。结果:未经内参较正的测定值,人、牛、兔的肌球蛋白重链(MHC)亚型及成肌调节因子(MRFs)基因表达均明显低于大鼠(P<0.01),提示各物种的同种基因存在差异性,其引物特异性较强;但经过内参校正后的测定值,人的MHC各亚型基因表达与大鼠含量较接近,有一定的交叉和基因相似性;而牛的含量虽可检出,但仍明显低于大鼠含量(P<0.01),表明其基因相似性较低;兔与大鼠基因未表现出相似性。提示,各物种同种基因的表达既存在差异性,又有一定的交叉和相似性。其蛋白水平的比较测定有待进一步研究。Objective: To compare the similarity of the gene expression of myosin heavy chain(MHC) isoforms and myogenic regulation factors(MRFs) in skeletal muscle among different animals by the primer of rat.Method: 24 samples of vastus lateralis respectively from rats,rabbits,bulls and human were fixed and homogenated with TRIZOL.Then the gene expression of MHC isoforms and MRFs in skeletal muscle among different animals was measured by realtime PCR method.Results:When not being calibrated by internal reference,the gene expressions of MHC isoforms and MRFs in skeletal muscles of rabbits,bulls and human were significantly lower than ones in rats(P0.01).This indicated that there are primer specialties among different animals.But when being calibrated by internal reference,the mRNA values of MHC isoforms in human were close to the ones in rat with the high similarity of gene.At same time,the mRNA values of MHC isoforms in bull were significantly lower than in rat(P0.01) with a low similarity of gene.On the other hand,there were no similarity of the genes between rabbit and rat. Indications: the expressions of the same gene of different species have differences,while they also share overlapping features and similarity.The comparison of the protein expression must be researched in next experiment.
