摘 要:通过土壤筛选和诱变,获得了一株对聚丁二酸丁二醇酯(PBS)具有降解作用的真菌Aspergillus versicolor DS0503-a,探讨了该菌株及其产生的PBS降解酶的作用机制,同时进行了菌株混合培养降解PBS的实验,从菌株互生和酶协同作用的角度探索了混菌降解PBS的特性和规律.结果表明:该菌株对PBS粉末和薄膜都具有明显的降解作用,产酶的最适pH值和温度分别为5.5和28℃;分泌的解聚酶以外切形式从羧基端对PBS链进行剪切,特定菌株的混合培养可以显著提高微生物对PBS的降解效率.Poly(butylenes succinate)(PBS)-degrading Aspergillus versicolor DS0503-a was obtained by ultraviolet light compound LiCl mutagenesis from the Aspergillus versicolor DS0503 which was isolated from soil.The optimum pH and temperature for the enzyme production of the strain was pH=5.5 and 28℃,respectively.The primary products after culture were identified by mass spectrometric analysis and the results indicated that the enzyme was exo-type and the cut the chain from the carboxyl end;affinity for the substrate was relative to the chain length of the carboxylic ester.Mixed strains were cultured to measure the PBS-degrading ability and the degrading activity of DS0401+DS0601 and DS0503-a+DS0601 strains mixture were increased observably.
