间接ELISA测定禽源大肠杆菌外膜蛋白抗体方法的建立

摘　　要：建立的检测禽源大肠杆菌外膜蛋白抗体的间接 Ｅ Ｌ Ｉ Ｓ Ａ，抗原最适包被质量浓度为 ５０ ｍ ｇ／ Ｌ；包被方式以 ４℃过夜为佳；待检血清作用时间为 １～１５ ｈ，酶标抗体作用时间为 １５ ｈ 时效果最好，酶标抗体最适稀释倍数为 １∶４ ０００。An indirect enzyme linked immunosorbant assay were developed for the detection of antibodies to outer membrane proteins (OMPs) of pathogenic Escherichia coli isolate O 18 566 from chicken origin. The results revealed that the optimal concentration for OMPs coating was 50 mg/L; the P/N for OMPs coated in 4℃ overnight was higher than which in 37℃ for 2 h; the optimal incubation time for the first sera (specific to OMPs developed in chickens) was 1 to 1.5 h; the optimal incubation time for the HRP conjugated rabbit IgG against chicken IgG was 1.5 h; the optimal dilution for HRP conjugated rabbit IgG was 1 ∶4 000.
