Successful Cryopreservation of Expanded Mouse Blastocysts by Vitrification after Being Fumigated with Liquid Nitrogen Gas

摘  要:采用液氮气熏蒸法,解决了玻璃化冷冻解冻过程中因急速降温和升温使胚胎遭受物理性损伤,导致透明带破裂而死亡这一关键性问题。结果在20或25℃室温下,使用EFS40或GFS40玻璃化溶液二步法冷冻保存的胚胎,解冻后透明带损伤率为0%,胚胎发育率近100%,囊胚孵化率超过80%,与对照组相比无显著性差异(P>0.05)。冷冻胚胎经移植后其妊娠率和产仔率(70%,60%和77%,72%)与对照组(86%和61%)相比无显著性差异(P>0.05)。Physical damage usually occured in mammalian embryos being rapidly frozen and thawed, evenly, the damage caused rupfor of zona pellucid (ZP) and death of the embryos, but it could be got rid of in this study. Expanded mouse blaStOCystSwere fulnigatdwith liquid nitrogen (LN2) gas for 2 min before they were cryopreserved in LN, by vitrification with EFS or GFS. The hozen embryos were thawed in warm water aamer being stayed for about 15 see in aiL the rates of development and hatChed blastocysts were almost 100 % and more than 80 % respectively, no damage of ZP was observed, the resultS didn't differ horn the contIDI (ho.05), the rates of pregnancies and full-term young were not different from the control either (M.05).
