摘 要:目的:探讨人巨细胞病毒(HCMV)对绒毛外细胞滋养细胞增殖功能的影响。方法:选择绒毛外细胞滋养细胞株HPT-8进行研究,体外培养时加入HCMV,免疫荧光法检测HCMV PP65,MTT法检测不同浓度HCMV、不同作用时间对HPT-8增殖的影响。结果:病毒感染组HPT-8细胞内可见大量红色HCMV PP65抗原信号;与正常对照组比较,10~100μl病毒液作用12 h HPT-8增殖均无异常(P>0.05),70μl和100μl病毒液作用24 h HPT-8增殖明显抑制(P<0.05),20、30、40、50μl病毒液作用48 h HPT-8增殖均明显抑制(P<0.05),10μl病毒液作用72 h HPT-8增殖明显抑制(P<0.05);其中,48 h和72 h时间点,20μl、30μl、40μl病毒液导致的细胞增殖异常差异无统计学意义(P>0.05)。结论:HCMV可感染HPT-8并在细胞内复制,抑制细胞增殖。Objective:To explore the effect of human cytomegalovirus(HCMV) on proliferative function of trophoblastic cells of external villus in vitro. Methods:Trophoblastic cell strain HPT-8 of external villus was selected and researched,HCMV was added in to the course of culture in vitro,immunofluorescence was used to detect HCMV PP65,MTT method was used to detect the effects of different doses of HCMV and different times on the proliferation of HPT-8. Results:In HCMV infection group,a large number of red HCMV PP65 antigen signals could be seen in HPT-8 cells;compared with normal control group,there was no significant difference in the proliferation rate among the HPT-8 cells treated with 10~100 μl HCMV fluid for 12 hours(P〉0.05);the proliferation of HPT-8 cells was inhibited obviously after treated with 70 μl and 100 μl HCMV fluid for 24 hours(P〈0.05);the proliferation of HPT-8 cells was inhibited obviously after treated with 20,30,40,50 μl HCMV fluid for 48 hours(P〈0.05);the proliferation of HPT-8 cells was inhibited obviously after treated with 10 μl HCMV fluid for 72 hours(P〈0.05);there was no significant difference in the proliferation rate among the HPT-8 cells treated with 20 μl,30 μl,40 μl HCMV fluid for 48 and 72 hours(P〉0.05). Conclusion:HCMV can infect HPT-8 cells and replicate in the cells,which may inhibit the proliferation of HPT-8 cells.
