摘 要:目的探讨CliniMACS系统进行CD^+34造血干细胞阳性富集的有效性、安全性及应用效果。方法对39例实体瘤和自身免疫性疾病患者动员后采集的外周血干细胞进行了CliniMACS系统阳性富集的CD^+34自体造血干细胞移植。结果阳性富集后CD^+34细胞纯度平均可达89.8%,CD^+34细胞平均回收率达78.9%,粒细胞-巨噬细胞集落形成单位(CFU—GM)数量为(15.0±7.0)×10^5/kg,阳性富集前、后细胞胎盘蓝拒染率比较差异无统计学意义,保持了细胞良好的生物学活性,移植后于2~3周内造血重建。结论应用CliniMACS系统进行CD^+34造血干细胞阳性富集,可安全、有效地应用于造血干细胞移植。Objective To investigate the validity and security of CD + 34 hematopoietic stem cells enrichment by using CliniMACS device. Methods Thirty -nine patients with solid tumor and autoimmune disease received CD+34 HSCT, the CD + 34 cells were enriched by CliniMACS device. Results After en- richment, the median purity of CD + 34 cells was 89.8%, the median recovery rate of CD + 34 cells was 78.9%, the granulocyte - macrophage cluster - colony forming unit ( CFU - GM) counts were ( 15.0±7.0)×105/kg. The refuse rate of typan blue was similar, compairing before and after enrichment, and the hematopoietic function recovers 2 -3 weeks after transplantation. Conclusions The feasibility and safety of the CD + 34 cells enrichment by using CliniMACS device in patient before HSCT were confirmed.
