摘  要:目的观察胰岛素样生长因子(IGF-1)对受损血管内皮细胞(VEC)增殖的影响。方法利用不同浓度(10、40、80、160 ng/ml)的IGF-1作用于血管内皮细胞48 h后,通过MTT法检测细胞的生长情况;采用划痕实验观察IGF-1对受损内皮细胞增殖的影响,并用PI染色观察细胞的情况,ELISA法测定VEGF含量。结果 10 ng/ml、20 ng/ml、40 ng/ml、80 ng/ml的IGF-1对血管内皮细胞的增殖都有不同程度的促进作用,IGF-1浓度为40 ng/ml、80 ng/ml的实验组与未加入IGF-1的对照组比较,差异有统计学意义(P<0.05),160 ng/ml的IGF-1对细胞增殖作用下降,但差异无统计学意义(P>0.05)。取40 ng/ml IGF-1作用于受损血管内皮细胞48 h后,细胞增殖速度较对照组明显加快,PI染色显示实验组死亡细胞明显减少,实验组培养液上清VEGF含量较对照组高,差异均有统计学意义(P<0.05)。结论 IGF-1对受损血管内皮细胞的增殖有促进作用,同时可减少其细胞凋亡,促进受损内皮细胞的修复,其机制可能与IGF-1促进细胞分泌VEGF有关。Objective To observe the effect of insulin-like growth factor-1(IGF-1) on proliferation and anti-apoptosis of injured vascular endothelial cell and investigate the mechanism.Methods Vascular endothelial cells cultured in vitro were treated with various concentrations(20,40,80,160 ng/ml)of IGF-1 for 48 h separately and determined by MTT method.The effect of IGF-1 on vascular endothelial cell was measured by cell migration experiment,then the number of dead cell was observed by PI stain and the level of VEGF was measured in culture media by ELISA.Results The concentrations of IGF-1(10,20,40,80 ng/ml) promoted the proliferation of vascular endothelial cell,the high concentration of IGF-1 inhibited the proliferation of vascular endothelial cell.The speed of 40 ng/ml IGF-1 promoting the proliferation of injured vascular endothelial cell was faster than that of control group and fewer apoptosis cells.Conclusions IGF-1 could promote the proliferation of vascular endothelial cell,and the effect might be VEGF-dependent.
