摘  要:以单体、致孔剂在不锈钢柱管中直接聚合的方法,合成了一种丙烯酰胺-甲基丙烯酸丁酯-N,N′-亚甲基双丙烯酰胺聚合物连续棒状疏水色谱柱.考察了聚合物单体中甲基丙烯酸丁酯的含量对于标准蛋白质的分离效果和介质疏水性的影响.用液相色谱中溶质的计量置换保留模型的四个线性参数对该色谱柱的色谱特性进行了表征.发现随着甲基丙烯酸丁酯比例的减小,柱疏水性增强,蛋白质对固定相亲合势增大.该色谱柱可用于蛋白质的分离纯化和保留机理的研究.Poly(acrylamide-co-butyl methacrylate-co-N, N'-methylenebisaciylamide ) hydrophobic interaction chromatographic (HIC) continuous rod column has been prepared in-situ by direct polymerization with monomer mixture and porogenic solvents in stainless steel tube. The effect of butyl methacrylate content in the monomer mixture on the separation efficiency and hydrophobicity of the synthesized polymers has also been investigated. The four linear parameters in the stoichiometic displacement model of retention could be used as the characterization parameters for this polymeric continuous rod column. It showed that the hydrophobicity of the continuous rod column would be increased as the content of butyl methacrylate decreased. Thus, the affinity of protein to the continuous HIC stationary phase increased. This column synsized in this paper could be used in separation and in the studies of retention mechanism of proteins.
