摘 要:为了解香蕉枯萎病菌(Fusarium oxysporum f.sp.cubense)1号生理小种(race 1)和4号生理小种(race4)丝裂原活化蛋白激酶(MAPK)基因focMK1,focMK4的序列差异,根据番茄枯萎病(Fusarium oxysporum f.sp.lycopersici)的MAPK基因相关序列设计引物,利用PCR和RT-PCR技术,分别克隆了香蕉枯萎病菌2个生理小种的基因序列和开放阅读框。结果表明,所获得的2个MAPK基因均含有3个内含子和4个外显子,2个基因序列只有一个碱基差异,编码氨基酸355个,氨基酸组成无差异;根据生物信息学软件预测编码蛋白没有信号肽,具有2个功能位点,分子量和pI分别为41ku和6.47。该基因编码的蛋白具有高度保守性,与大多数植物致病菌的同源性都在90%以上,不同病原镰刀菌间的同源性接近100%。Vascular wilt of bananas is one of the most serious banana disease worldwide.The MAP kinase is a signal transducer,plays an important role in pathogenesis mechanism.According to the homologous gene sequence documented in Genbank,a pair of primers were designed.Using PCR and RT-PCR,the gene and open reading frame(ORF) from two races of Fusarium oxysporum f.sp.cubense were amplified respectively.The gene included 3 introns and 4 exons.The ORF encoded 355-amino acid polypeptide with 41 ku of calculated molecular weight and 6.47 of pI.The protein had a high evolutionary conservative,shared homology of more than 90% with many fungal plant pathogens and there was no difference between the two races of Fusarium oxysporum f.sp.cubense in amino acid sequences.
