摘 要:以芘(pyrene)为荧光探针,应用稳态荧光法探讨阳离子型Gemini表面活性剂G14-3-14与脱氧核糖核酸(DNA)的相互作用,并求得G14-3-14的临界胶束浓度(CMC)为3.8×10^-4mol.L^-1.根据DNA对G14-3-14370nm处共振光散射信号的增强效应,建立以G14-3-14为探针测定DNA的共振光散射法,并对介质酸度、离子强度、表面活性剂浓度及共存物质干扰等实验条件进行优化.在最佳条件下绘制了工作曲线,该方法线性范围为0-3 450μg.L^-1,检出限为28.6μg.L^-1,用于合成样品中DNA的测定,结果令人满意.此外,通过红外光谱及共振光散射光谱初步探讨了G14-3-14与DNA相互作用的机理,即G14-3-14在DNA分子表面进行聚集和长距组装,从而导致G14-3-14共振光散射信号的增强,这也是该方法测定DNA含量的理论基础.The paper investigated the interaction between a new Gemini surfactant G14-3-14 and DNA by applying pyrene as fluorescence reagent.A detection assay was proposed based on the enhanced resonance light scattering(RLS) signals of G14-3-14 at 370 nm by DNA at pH 3.51.The effects of pH,ionic strength,G14-3-14 concentration and coexisting substances on the RLS enhancement were discussed,respectively.The intensity of RLS signal was proportional to the concentration of DNA in the range from 0 to 3 450 μg·L-1 with the determination limit(3σ) of 28.6 μg·L-1.This novel method had been applied to the assay for four synthetic samples with satisfactory results.Infra-red and RLS spectra were used to study the interaction mechanism.The long range assembly of G14-3-14 on the molecular surfaces of DNA occurred with electrostatic and hydrophobic interaction,which results in enhanced resonance light scattering(RLS) signals.
