摘 要:目的观察核结合因子α亚单位1(Cbfα1)在糖尿病肾病(DN)大鼠肾实质内小动脉上的表达及其变化规律,探讨其对DN血管病变的影响。方法设对照组和DN组两组,建立链脲佐菌素(STZ)诱导的DN大鼠模型。茜素红染色观察肾实质内小动脉周围钙盐沉积,并分别采用免疫组化及EnRNA原位杂交法检测Cbfα1在肾实质内小动脉上的蛋白表达及基因定位。结果(1)茜素红染色DN组第4~12周时偶见肾内小动脉少许点状红色钙盐沉积,第24周时可见肾实质内小动脉及周围组织有较多红色点片状沉积;对照组茜素红染色为阴性。(2)DN组肾实质内小动脉Cbfα1蛋白及mRNA在4w时已有表达,随时间延长表达逐渐增强,各时间点均明显高于对照组(P〈0.01);对照组各时间点表达差异无统计学意义(P〉0.05)。结论Cbfα1是糖尿病血管钙化的重要转录因子及早期生物学标志;肾内小动脉上Cbfα1的表达变化可能参与DN进展。Objective To observe the expression of core-binding factor α1 (Cbf α1 ) in renal arteriole and explore the effect on vascular lesion of diabetic nephropathy (DN). Methods Adult SD male rats were divided into diabetic and control groups. Paraffin sections of renal tissue of rats at the same time points were stained by routine methods and alizarin red, immunohistochemistry, hybridization in situ. Results The a bit calcium salt deposited in renal arteriole at 4 week and a lot calcium salt at 24 week. From the 4th week, the number of Cbf α1 staining and mRNA in diabetic renal arteriole was significantly increased compared with that of control group. Conclusions Cbf α1 has already expressed in renal arteriole before the formation of vascular calcification. Before vascular calcification, Cbf α1 in diabetic renal arteriole is significantly upregulated, indicating that Cbf α1 is a key regulating factor of diabetic vascular calcification. Alteration of the expression of Cbf α1 in renal arteriole may participate in the development of DN.
