摘 要:目的用microRNA-21抑制剂转染Flk1+间充质干细胞(MSC),探讨microRNA-21对Flk1+MSC(MSC)迁移的影响。方法在脂质体介导下用microRNA-21抑制剂瞬时转染Flk1+MSC,用Transwell技术检测细胞迁移,用Real-time PCR技术检测micro-RNA的表达。结果与对照组相比,实验组中microRNA-21表达明显下调,实验组细胞在12、16和20 h迁移率分别是对照组的77.5%、71.3%和69.8%。结论 microRNA-21可调控Flk1+MSC的迁移。Objective To evaluate the effect of microRNA-21 on migration of Flk1+ mesenchymal stem cells(MSCs).Methods Flk1 +MSCs were Transfected with microRNA-21 inhibitor using Lipofectamin2000 and their migratory activity was evaluated by Transwell.Real-time PCR was applied to detect expression of microRNA.Results Compared to control group,the expression of microRNA-21 was significantly reduced in the inhibitor-transfected group.Moreover,the number of cells migrated through Transwell in the inhibitor-transfected group was 77.5%,71.3% and 69.8% of that in the control group at 12,16 and 20 h,respectively.Conclusion microRNA-21 can regulate the migratory capacity of Flk1+MSCs.
