摘 要:目的:打破小菘菜种子休眠,加速转育进程。方法:以小菘菜06 s 112回交代种子为试验材料,利用幼胚培养技术打破种子休眠。结果:幼胚以"由透明向不透明过渡"的鲜嫩幼胚即授粉后约35 d的种子幼胚为宜;促进萌发的培养基是MS+0.8%琼脂+3%蔗糖+1.5 mg/L 6-BA+0.5 g谷氨酰胺,一般7 d幼胚即可萌发,发芽率达90%以上;播于穴盘后成苗率为95%以上;植株外部形态和花期育性完全正常。结论:该方法可促使休眠的种子提前萌发,在核基因雄性不育系的转育过程中,可提前种子播期30 d左右,加速了转育进程。[Objective]In order to accelerate the process of male sterile line breeding,probe a method to break seeds dormancy of Komatsuna.[Method]with Komatsuna 06 s 112 seed got in back-crossing generation as material,and by embryo culture technology to break seed dormancy.[Result]The suitable embryo was about 35 days after pollination "from transparent to opaque".And the proper culture medium was MS+0.8% agar+3% sucrose+1.5 mg/L 6-BA+0.5 g glutamine,which could promote germination.Usually,the embryo germinated in one week,and the germination rate could up to 95%.The external features of plants and fertility of flowers were perfectly normal.[Conclusion]The aboved method could break seeds dormancy and promote seed to germinate in advance.In the process of male sterile line breeding,the transfer process could accelerated obviously if the sowing time was set forward for about 30 days.
